Instructions:

1. Install the provided pressure regulator on the CO2 cylinder. Ensure there is no air leakage at the connection. Do not open the cylinder yet. Connect the output connector of the pressure regulator to the hose on the CO2 intake pipe located on the back of the CO2 box, and secure it with a compression ring to prevent air leaks.

2. Wipe the working chamber of the CO2 box with alcohol. Turn on the UV lamp power switch on the back of the box and disinfect for 1–2 hours.

3. Connect the faucet to the upper left side of the tank using a water pipe. The water-jacketed CO2 tank operates in one step.

4. Turn on the power and set the power switch to "1". The green indicator light should be on.

The water-jacketed CO2 tank must be filled with water first. Open the faucet and slowly add water into the tank. As the water level rises, wait for 3–5 seconds once the water level indicator turns off. At this point, the water level should be between the low and high marks, and the water level light should not be bright. The equipment can now be used.

Adjust the temperature setting to your desired value (e.g., 37°C). The heating indicator light (green) should be on, indicating that the system is heating. (Note: If the ambient temperature exceeds 25°C, consider using an air conditioner to cool the environment.)

Set the CO2 control switch to "0".

Once the temperature reaches the set value, open the CO2 cylinder. Before opening, make sure the pressure regulator is loosened as much as possible to avoid excessive pressure that could cause the rubber tube to burst. The pressure gauge on the regulator shows the internal pressure of the CO2 cylinder. Slowly turn the relief valve knob clockwise until the pressure output reads 0.05 MPa, with the pointer in the middle of the scale. Use the dial to set the CO2 concentration (factory preset to 5%). Before turning on the CO2 control switch, ensure the air flow meter and CO2 flow meter are turned off. Once the CO2 controller displays “1”, CO2 will enter the inner chamber. As the concentration increases, the display will show the actual CO2 level. When the set value is reached, the device automatically switches to the air and CO2 mode. At this time, adjust the air flow meter to 760ml/min (pre-set at the factory) and the CO2 flow meter to 40ml/min (also pre-set at the factory). If the float deviates, adjust the knobs accordingly.

When the temperature stabilizes within ±0.5°C and the CO2 concentration is reached, you can begin cell culture. Place a water tray inside to maintain proper humidity.

This machine should be used according to the above instructions after the first use or long-term storage. It is recommended to perform a contamination check inside the box before starting any formal cultivation.

5. Shutdown Procedure

After completing the culture, remove the culture dish from the box and reinsert any unused dishes (check if distilled water in the water tray needs to be replenished). If the culture is being stopped, close the CO2 cylinder first, then turn off the air and CO2 flow meters, and set the CO2 intake switch to “0”. Finally, turn off the main power. If the machine will not be used for a long time, remove the water tray and dry the working chamber. Turn on the CO2 box at 37°C for 2 hours to dry the interior, then cut off the power supply.

6. Temperature Adjustment Method

When the machine leaves the factory, it is set to 37°C. If you notice heavy condensation on the glass door during winter, making it hard to see inside, adjust the temperature knob located behind the CO2 box. Turning the knob to the left increases the temperature, while turning it to the right decreases it. Adjust until the contents of the box are clearly visible. In summer, if the actual temperature inside the box is higher than the set value, turn the knob to the right to lower the temperature until it reaches 37°C.

7. Fan Adjustment Method

During operation, the fan speed affects both the temperature uniformity and the control accuracy. If the airflow is too strong and interferes with the cultured items, adjust the fan speed knob on the back of the CO2 box until the desired airflow is achieved.

Precautions

1. This machine should be placed in a clean, well-ventilated room away from direct sunlight, strong electromagnetic fields, and heat sources. To ensure accurate temperature control, it is recommended to operate the CO2 box in an environment between 15°C and 25°C.

2. Always loosen the pressure regulator before opening the CO2 cylinder to prevent the gas hose from bursting.

3. If the difference between the ambient temperature and the set temperature is less than (RT + 5°C), use an air conditioner to reduce the ambient temperature. During the entire culture process, the ambient temperature should remain stable, otherwise it may affect the internal temperature control of the CO2 box.

4. Monitor the pressure regulator output and the flow rates of both flow meters regularly. Avoid adjusting any valves inside the control box arbitrarily. If the CO2 concentration is inaccurate, identify the root cause before making any adjustments.

5. Replace the CO2 cylinder when the pressure drops below 1MPa. When replacing, close the cylinder valve first, loosen the pressure regulator, remove it, and install it on the new cylinder.

6. If the inner door of the CO2 box becomes stuck, turn off the CO2 control switch first. After closing the door, wait until the CO2 concentration display shows “0.0” before turning the switch back on.

7. Ensure the CO2 system has a good grounding device for safety.
