Researchers have discovered that zinc can "starve" bacteria by blocking the uptake of manganese in *Streptococcus pneumoniae*, a breakthrough that could lead to new antibacterial treatments. Previously, scientists knew that zinc played a role in the body's defense against infections, but this study reveals the exact mechanism. The research shows that zinc interferes with a key transporter protein (PsaBCA) that allows *S. pneumoniae* to absorb manganese, an essential metal for its survival and infection process. The PsaBCA transporter uses a unique "spring-hammer" mechanism to bind metals. Due to the size difference between manganese and zinc, the transporter binds them differently. Zinc, being smaller, causes the transporter to over-clamp, creating a structural block that prevents it from properly absorbing manganese. Without manganese, the bacteria lose their ability to invade and cause disease. This discovery spans multiple scientific disciplines, including chemistry, biochemistry, microbiology, and immunology. It provides detailed insights into how the transporter functions at the atomic level and how zinc can be used to disrupt bacterial activity. The findings suggest that without manganese, these bacteria become vulnerable to the immune system. For the first time, scientists have fully understood the functional mechanisms of this type of transporter. With this knowledge, researchers can now work on developing next-generation antimicrobials that specifically target and block these transporters, offering new hope in the fight against antibiotic-resistant strains.
